Methodology

Hexokinase-G-6-PDH

Useful as the principle tool for diagnosing glucose tolerance in the evaluation of diabetes mellitus, hypoglycemia, and related disease conditions.

Specimen Requirements

Patient Preparation
Patient should have nothing to eat or drink for at least 8 to 12 hours before test (except for sips of water). Patient should remain fasting and abstain from smoking, chewing gum, or LifeSavers® during testing period. Patient may not leave during testing period.

 

After drawing fasting specimen, administer glucose tolerance solution as follows:

 

Children (by weight)
<20 pounds(9.1 kg): consult  physician

20-24 pounds(9.1-10.9kg): 1.6 ounces

25-29 pounds(11-13.2kg): 2.0 ounces

30-34 pounds(13.3-15.4kg): 2.4 ounces

35-39 pounds(15.5-17.7kg): 2.8 ounces

40-44 pounds(17.8-20kg): 3.2 ounces

45-49 pounds(20.1-22.2kg): 3.6 ounces

50-54 pounds(22.3-24.5kg): 4.0 ounces

55-59 pounds(24.6-26.8kg): 4.4 ounces

60-64 pounds(26.9-29kg): 4.8 ounces

65-69 pounds(29.1-31.3kg): 5.2 ounces

70-74 pounds(31.4-33.6kg): 5.6 ounces

75-79 pounds(33.7-35.8kg): 6.0 ounces

Specimen Collection

Submit only 1 of the following specimens:

 

Plasma

Container/Tube: Mint-top (lithium heparin) tube

Specimen Volume: 1 mL of plasma for each timed specimen

Collection Instructions: Fasting patient (8-12 hours)

1. After drawing a fasting specimen, give a measured amount of chilled glucose tolerance solution before drawing remaining specimens (patient must drink entire beverage within 5 minutes).

2. Draw specimens at 1-, 2-, and 3-hours for duration of test.

Note: 1. Indicate plasma and draw times.

2. Label specimen appropriately (phlebotomist’s initials, corresponding draw time, and as plasma).

 

Serum

Container/Tube: Serum gel tube

Specimen Volume: 1 mL of serum for each timed specimen

Collection Instructions: Fasting patient (8-12 hours)

1. After drawing a fasting specimen, give a measured amount of chilled glucose tolerance solution before drawing remaining specimens (patient must drink entire beverage within 5 minutes).

2. Draw specimens at 1-, 2-, and 3-hours for duration of test.
